Microplitis bicoloratus Bracovirus Promotes Cyclophilin D-Acetylation at Lysine 125 That Correlates with Apoptosis during Insect Immunosuppression

چکیده

Cyclophilin D (CypD) is regulated during the innate immune response of insects. However, mechanism by which CypD activated under immunosuppression not understood. Microplitis bicoloratus bracovirus (MbBV), a symbiotic virus in parasitoid wasp, bicoloratus, suppresses immunity parasitized Spodoptera litura. Here, we demonstrate that MbBV promotes acetylation S. litura, resulting an immunosuppressive phenotype characterized increased apoptosis hemocytes and MbBV-infected cells. Under infection, inhibition significantly rescued apoptotic cells induced MbBV, point-mutant fusion proteins CypDK125R-V5 were deacetylated. The CypD-V5 acetylated Deacetylation can also suppress MbBV-induced increase apoptosis. These results indicate involved MbBV-suppressed via CypD-acetylation pathway litura on K125.

Disruption effect of Microplitis bicoloratus polydnavirus EGF-like protein, MbCRP, on actin cytoskeleton in lepidopteran insect hemocytes.

Microplitis bicoloratus is a braconid endoparasitic wasp associated with the polydnavirus named Microplitis bicoloratus bracovirus (MbBV). Parasitism of Spodoptera litura larvae leads to an impaired cellular immune response and to the disappearance of the 42 kDa actin in host hemocytes. In this work, we investigated if the absence of actin in blood cells was related to MbBV infection. An MbBV g...

PTP-H2 and PTP-H3 from Microplitis demolitor Bracovirus localize to focal adhesions and are antiphagocytic in insect immune cells.

Viruses in the family Polydnaviridae are symbiotically associated with parasitoid wasps. Wasps inject polydnaviruses (PDVs) when laying an egg into their insect host, and expression of viral gene products causes several physiological alterations, including immunosuppression, that allow the wasp's progeny to develop. As with other PDVs, most Microplitis demolitor bracovirus (MdBV) genes are rela...

A Transcriptome Analysis Suggests Apoptosis-Related Signaling Pathways in Hemocytes of Spodoptera litura After Parasitization by Microplitis bicoloratus

Microplitis bicoloratus parasitism induction of apoptotic DNA fragmentation of host Spodoptera litura hemocytes has been reported. However, how M. bicoloratus parasitism regulates the host signaling pathways to induce DNA fragmentation during apoptosis remains unclear. To address this question, we performed a new RNAseq-based comparative analysis of the hemocytes transcriptomes of non-parasitiz...

Glucose Stimulates GAPDH Acetylation at Lysine 254

The altered metabolism in most tumor cells consists of elevated glucose uptake and increased glycolysis even in the presence of high oxygen tension. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is an obligatory enzyme in glycolysis. Here, we report that acetylation at lysine 254 (K254) increases GAPDH activity in response to glucose. Furthermore, acetylation of GAPDH (K254) is reversibly re...
